Yes, absolutely! In fact, most puzzles require you to do this. You often need to use a fairy to open one door, walk your characters through it, and then drag the fairy out of that lantern and into a new one to open the next door.

Do I need a third person to control the mouse?

No, you don't need three people! Fireboy and Watergirl 6 is still a 2-player game. One of the players (usually the one playing on the side of the keyboard closer to the mouse) can quickly grab the mouse to move the fairies while their character is standing safely.
